Veterinary diagnostic laboratories are constantly required to perform Antibiotic Susceptibility Testing with animal samples. ETEST® is the ideal manual method for routine AST operations.

 

It consists of a predefined gradient of antibiotic concentrations placed on a plastic strip, which are used to determine the Minimum Inhibitory Concentration (MIC) of antibiotics.
The well-established ETEST
® method demonstrates high performance in AST, as it provides a level of precision that automated methods cannot reach.

A vacuum pen that simplifies the application of Etest® strips. The applicator is held like a pen and the evacuation hole covered with the finger tip to create suction.  The suction cup is placed on the strip to lift it up and then positioned onto the agar surface. The applicator tray makes it easy to set up Etest panels containing multiple antimicrobials per plate. Simple place the antimicrobial strips of choice into the wells as shown below, and use a manual applicator device such as the Mini Grip-It to place the strips on to agar plates.

The Mini Grip-It is a device that makes it easier to grab, hold, and move small and light objects from one place to another. The suction cup picks up hard to handle items such as Etest strips, antibiotic disks, and glass/plastic cover slips. The whole apparatus can be autoclaved.

Etest is susceptible to moisture and open strips must be stored with active desiccant. When using alternative storage units, it is mandatory to ensure that desiccant capsules are used with an air-tight container.  As desiccant absorbs moisture it will change color from yellow/orange to white. Capsules are reactivated by heating for 2 hours at 70°C or until the orange color is restored.

This unique ETEST® gradient strip is designed for phenotypic detection of Enterobacteriaceae possessing MBLs (metallo ß-lactamase), including strains with the recently discovered NDM-1 resistance gene. This type of bacterium is resistant to nearly all ß-lactam antibiotics, and they are not inhibited by clinically available ß-lactamase inhibitors. The MBL MP/MPI strip is specifically designed to detect these problematic resistance phenotypes by measuring the inhibitory concentration (IC) ratio of meropenem to meropenem + EDTA to detect MBLs. The IC values serve to measure a ratio (MP/MPI), and cannot be used to determine MIC values for MP.

ETEST® MBL MP/MPI is a double-sided strip consisting of Meropenem (MP) on one side and Meropenem plus EDTA (MPI) on the other:

. MP concentration: 0.125 –8 μg/ml

. MPI concentration: 0.032 –2 μg/ml plus constant level of EDTA

The presence of MBL is indicated by a reduction of MP IC by ≥3 doubling dilutions in the presence of EDTA (i.e. IC ratio of MP/MPI of ≥8), the appearance of phantom zones, or deformation of ellipses (see fig 1).  The table below provides examples of proper Etest MBL MP/MPI interpretation.

The image above shows an isolate with a MPI IC of 0.032 and an MP IC of ≥8, therefore the MP/MPI ratio is >250 indicating an MBL positive strain.
